RESEARCH INTERESTS
Dr. Fan specializes in investigating the pathogenic genes of rare/complex diseases and identifying biomarkers for oral cancer, with a particular focus on understanding the genetic basis of disease inheritance and susceptibility, including rare diseases and cancer genetics. Their study involved mutational profiling of OCSCC tumors in 165 advanced patients, revealing frequent mutations in 10 oncogenic pathways, such as TP53, FAT1, and NOTCH1. Certain subgroups with specific alterations showed a negative impact on survival, indicating their clinical significance. The identification of multiple targetable mutations across various pathways suggests the potential for novel therapeutic approaches. Additionally, Dr. Fan explored the contribution of primary immunodeficiency disorder (PID)-associated genetic variants to childhood-onset systemic lupus erythematosus (cSLE), a condition characterized by a dysregulated immune response. Targeted sequencing identified damaging PID variants in 30.5% of cSLE patients, indicating their potential role in cSLE susceptibility. The study observed both dominant and recessive traits, along with higher mutation loads in cSLE patients compared to controls. These findings highlight the involvement of rare damaging variants in PID-related genes in the development of cSLE.
